A UV-spectrophotometric and RP-HPLC method was developed and validated for concurrent estimation of fexofenadine hydrochloride (FEX), montelukast sodium (MLK) and ambroxol hydrochloride (AMB). In RP-HPLC chromatography, the retention time for FEX, MLK and AMB was 2.775 min, 9.529 min and 5.261 min, respectively and detection at λmax 210 nm for all three of drugs (overlain spectra). Linear regression analysis shows a good linear relationship; in the concentration range of 10-30, 10-50 and 5-25 μg/mL; for FEX, MLK and AMB with r2 > 0.999. The UV-spectrophotometric estimation was done by two methods; zero order and first derivative method. These methods were based on the measurement of absorbance at 220.20, 283.42 and 245.84 nm in zero order; 221, 287.52 and 262.25 nm in first order derivative method. In both UV methods, the drugs follow the Beer-Lambert’s law in the concentration range of 6-26, 4-24 and 8-28 μg/mL, respectively showing correlation coefficient r2 > 0.999. The RP-HPLC and UV methods showed good reproducibility and recovery with the percent relative standard deviation (RSD) less than 5%. These methods were successfully applied for reliable quantification of FEX, MLK and AMB in a commercial and routine analysis in combined dosage form
